A second child has died following a horror 4WD rollover that left a seven-year-old girl dead and five other children injured.

Two children have died following a single-vehicle crash west of Melbourne near Bacchus Marsh.

A second child has died following a horror rollover west of Melbourne that has now claimed the lives of a seven-year-old girl and an 11-year-old boy.

Emergency services were called to the single-vehicle crash near the intersection of Nerowie and Parwan-Exford roads at Parwan just before 4pm on Tuesday.

Police said a 34-year-old woman was driving an eight-seater 4WD carrying seven children when the vehicle rolled and came to a stop on grass between the road and a nearby paddock.

Paramedics desperately worked to save a seven-year-old girl, but she died at the scene.

An 11-year-old boy was airlifted to hospital in a critical condition following the crash.

Victoria Police confirmed on Wednesday morning that the boy had died in hospital overnight, becoming the second child to lose their life in the tragedy.

Five other children who were travelling in the vehicle were taken to hospital with non-life-threatening injuries.

The 34-year-old driver was also taken to hospital for observation.

Major Collision Investigation Unit detectives are now investigating the circumstances surrounding the rollover and are working to determine what caused the vehicle to leave the road.

Police are appealing for anyone who witnessed the crash or may have CCTV or dashcam footage of the vehicle or incident to come forward.
