A pedestrian has been killed and a woman injured after a car ploughed into a medical centre in Melbourne’s west.

Emergency services were called to the busy intersection of Keilor Road and Hoffmans Road in Niddrie around 9.30am on Monday after a car lost control and crashed into a doctor’s office.

A male pedestrian died at the scene, while a woman inside the building was also struck and airlifted to hospital with serious injuries to her upper and lower body.

The 63-year-old male driver of the car has been taken to hospital, where he remains under police guard.

“The investigation into the circumstances surrounding the collision remains ongoing,” Victoria Police said in a statement.

Locals are shaken by the tragedy. A witness told 7NEWS that the “screams were just horrendous”.

“We just heard a big crash and a big thump...definitely knew something bad had happened.”

Motorists are also being urged to avoid the area.
