The bus is believed to have been overtaking another vehicle when the crash occurred, Victoria Police Sergeant Mark McDonald said.

Two people died at the scene.

The male driver and one passenger sustained critical injuries, with one taken to Mildura Hospital and the other airlifted to The Alfred in Melbourne.

Three other passengers suffered serious injuries, while another four were taken to Mildura Base Hospital for observation.

Police told 7NEWS that all the passengers were employed at nearby farms, and that many of them were from Samoa.

“It’s difficult to see why it’s happened,” McDonald said.

“Obviously, the weather’s fine, road conditions appear to be quite good, and there’s no indication there were any animals or anything like that involved.”

The Calder Hwy between Kulkyn Way and Brownport Rd was expected to be closed for several hours.

Police are investigating how the crash happened and anyone with information is urged to contact them.
